How Long Does Daridorexant Take to Work? What to Expect

If you've started treatment for long-term insomnia, one question matters more than almost anything else:

“When should I expect my sleep to improve?”

With daridorexant, the answer isn't necessarily one dramatic night's sleep.

Does daridorexant work on the first night?

Daridorexant acts on the brain's orexin system.

Orexin promotes wakefulness. Daridorexant blocks orexin receptors, reducing the wake drive and allowing sleep to occur.

The medicine acts after a nightly dose, but your experience of improvement may build over time.

Clinical trials measured improvements after one month and three months, while patient-facing providers report that some people notice changes during the first nights or weeks.

What improvements should I look for?

Don't judge treatment only by:

“Did I fall asleep instantly?”

Chronic insomnia can involve several problems:

  • taking too long to fall asleep
  • repeatedly waking during the night
  • waking too early
  • insufficient total sleep
  • tiredness and poor concentration during the day.

Daridorexant has been studied across both night-time sleep and daytime functioning.

You may therefore notice improvement as:

falling asleep more easily → spending less time awake during the night → getting more total sleep → functioning better during the day.

What if it isn't working after a few nights?

Don't immediately assume treatment has failed.

Check whether you are:

  • taking it consistently as prescribed
  • taking it within 30 minutes before bed
  • avoiding taking it immediately after a large meal
  • maintaining good sleep habits.

A large meal shortly before daridorexant can delay its effect on sleep onset.

What if it still isn't helping?

Treatment shouldn't continue indefinitely without benefit.

NICE recommends assessing response within three months, stopping daridorexant if long-term insomnia has not responded adequately, and reviewing continued treatment periodically thereafter.

If your sleep remains poor, a clinician may need to reconsider the diagnosis as well as the medication.

Conditions such as sleep apnoea, restless legs, anxiety, depression, pain, menopause symptoms or other medicines can all influence sleep.

How Pharmulous can help

At Pharmulous, we don't view insomnia treatment simply as:

“Tablet prescribed — job done.”

We review your online assessment and look at whether you are falling asleep better, staying asleep longer and, importantly , whether poor sleep is still affecting your daytime life.

If treatment isn't achieving its intended purpose, it deserves review.
