- CRB-913: What Do the New Weight-Loss...

Another once-daily weight-loss tablet has entered the obesity-research spotlight but this one works very differently from Wegovy tablets and Foundayo.
CRB-913 is an experimental oral medicine being developed for obesity. New early-stage trial results released in September 2026 suggest it produced meaningful weight loss over 12 weeks.
But CRB-913 is still at a very early stage of development, so the results need some context.
What is CRB-913?
CRB-913 is a CB1 receptor inverse agonist.
That makes it fundamentally different from medicines such as semaglutide and orforglipron, which act on the GLP-1 receptor.
CB1 receptors form part of the body's endocannabinoid system and are involved in appetite, metabolism and energy balance.
Earlier medicines targeting this pathway caused concerns because they also affected receptors in the brain and were associated with psychiatric side effects.
CRB-913 has therefore been designed to be peripherally restricted, meaning its developers are attempting to limit how much of the medicine reaches the central nervous system.
What did the new CANYON-1 trial find?
CANYON-1 was a Phase 1b study involving 254 adults with obesity who did not have diabetes.
Participants received placebo or once-daily CRB-913 at doses of 20 mg, 40 mg or 60 mg.
After 12 weeks, average weight reductions were:
- 20 mg: 2.8%
- 40 mg: 3.3%
- 60 mg: 5.0%
- Placebo: approximately 0%
All three doses produced statistically significant weight reduction compared with placebo. Among people who completed treatment at 60 mg, 44.4% lost at least 5% of their starting body weight.
Does CRB-913 cause fewer stomach side effects than GLP-1 medicines?
This is where the headlines need caution.
Corbus reported relatively low rates of vomiting and constipation and suggested CRB-913 may have a favourable gastrointestinal tolerability profile.
However, CRB-913 has not been compared directly with Wegovy tablets, Foundayo, Mounjaro or other established weight-management medicines.
The comparisons released by the company use results from separate trials conducted over different durations and in different populations.
So it is too early to say that CRB-913 causes fewer side effects than GLP-1 treatments.
Is CRB-913 available in the UK?
No.
CRB-913 is an investigational medicine and has not been authorised by the MHRA.
These are Phase 1b topline results rather than completed Phase 3 evidence. A larger Phase 2 study is expected to begin in 2027, and substantially more evidence will be needed before regulators can decide whether CRB-913 is sufficiently safe and effective for routine use.
It should therefore not be confused with the oral weight-management treatments already authorised in the UK.
Why is this research interesting?
The significance of CRB-913 is not simply that it is another tablet.
It represents a potentially different non-incretin approach to obesity treatment.
If later studies confirm the early findings, medicines working through different biological pathways could eventually expand the options available to people who do not tolerate, respond adequately to, or wish to use existing treatments.
But Phase 1 results are a starting point not proof of long-term effectiveness or safety.
How Pharmulous can help
Pharmulous follows emerging obesity research closely, while basing patient care on currently authorised treatments and established clinical evidence.
Our clinicians assess each patient's medical history, previous treatment, response, tolerability, weight trajectory and longer-term goals rather than assuming one medicine is right for everyone.
If you are considering medical weight management, including a tablet-based approach or longer-term weight maintenance, the Pharmulous clinical team can review the currently available and clinically appropriate options with you.
